چکیده

Aspherical magnetic particles orient in a magnetic field due to magnetic shape anisotropy. They also emit different fluxes of light from their different geometric faces due to self-absorption and total internal reflection within the particles. The particles rotate in response to rotating magnetic fields and appear to blink as they rotate. We have made pancake and chain shaped particles and magnetically modulated their fluorescent intensities. Demodulating the signal extracts the probe fluorescence from electronic and optical backgrounds dramatically increasing signal to noise ratios. The probes have applications in sensitive and rapid immunoassays, improved intracellular sensors, and inexpensive single molecule analysis. © 2003 American Institute of Physics. @DOI: 10.1063/1.1556926#

Magnetically modulated optical nanoprobes

We have developed magnetically modulated optical nanoprobes ~MagMOONs! to magnetically modulate the signal from fluorescent probes and thus separate it from autofluorescence, electronic offsets, and other background signals. These microand nanosized particles emit fluorescence signals, indicating chemical concentrations, and blink in response to rotating magnetic fields. Demodulating the signal...

Magnetically modulated fluorescent probes in turbid media

Clemson University Department of Chemistry, 371 Hunter Laboratories, Clemson, SC. 29634 * Corresponding author: [email protected] Abstract Magnetically modulated optical nanoprobes (MagMOONs) were used to detect and distinguish probe fluorescence from autofluorescent backgrounds in turbid media. MagMOONs are micro/nano-sized particles with magnetically controlled orientation and orientation-de...

Tracking Anisotropic Optical Tracers to Study Biophysical Processes and Cytotoxicity

We use anisotropic optical tracers (also called magnetically modulated optical nanoprobes – MagMOONs or MOONs for non-magnetic nanoprobes in this dissertation) to study biophysical processes such as enzyme-catalyzed cleavage through tissue, intracellular transport of these tracers and cytotoxicity based on this transport. The anisotropic optical properties cause these tracers to blink when rota...

Eu, Gd-Codoped Yttria Nanoprobes for Optical and T₁-Weighted Magnetic Resonance Imaging.

Nanoprobes with multimodal functionality have attracted significant interest recently because of their potential applications in nanomedicine. This paper reports the successful development of lanthanide-doped Y₂O₃ nanoprobes for potential applications in optical and magnetic resonance (MR) imaging. The morphology, structural, and optical properties of these nanoprobes were characterized by tran...

Microrheology with modulated optical nanoprobes (MOONs)

Metal-capping of one hemisphere of a nano or microparticle breaks the particle’s optical symmetry, allowing its orientation to be tracked using fluorescence and reflection. Tracking orientation and rotation allows the torques acting on the particles be inferred. In addition to serving as rotational nanoviscometers these probes report on local Brownian, magnetic and biomechanical torques. r 2005...
